DONEP 10 MG TABLET

DONEP 10 MG TABLET, manufactured by ALKEM LABORATORIES LTD., is a medication used in the management of Alzheimer's disease.

  • Composition Each tablet contains Donepezil Hydrochloride 10 mg.

  • Uses DONEP 10 MG TABLET is prescribed for the treatment of mild, moderate, and severe dementia associated with Alzheimer's disease. It functions by increasing the levels of acetylcholine, a natural chemical in the brain essential for memory and learning, thereby helping to improve cognitive functions such as memory, attention, awareness, and the ability to perform daily tasks. It does not provide a cure for Alzheimer's disease but can help slow the worsening of symptoms.

  • Dosage and Administration The typical starting dose is 5 mg once daily, taken in the evening just before bedtime. After 4 to 6 weeks, the dosage may be increased to 10 mg once daily, as directed by a healthcare professional. For moderate to severe Alzheimer's, a dose of 23 mg once daily may be considered after at least three months on 10 mg daily. The tablet can be taken with or without food.

  • Side Effects

    • Common Side Effects: Nausea, vomiting, diarrhea, loss of appetite, weight loss, muscle cramps, fatigue, headache, dizziness, and trouble sleeping.
    • Serious Side Effects: Fainting, slow heartbeat, chest pain, new or worsening breathing difficulties, severe stomach pain (potentially indicating an ulcer), black or tarry stools, bloody vomit (signs of gastrointestinal bleeding), seizures, and allergic reactions (rash, swelling).
  • Precautions Inform your doctor about any history of heart problems (including irregular or slow heartbeat), stomach ulcers or bleeding, epilepsy or seizures, asthma or other lung diseases (like COPD), liver disease, or difficulty urinating. DONEP 10 MG TABLET may interact with certain medications, including NSAIDs, anticholinergics, and anesthetics, which could increase the risk of side effects. Alcohol consumption should be avoided. This medicine may cause dizziness or drowsiness; exercise caution when driving or operating machinery.

  • Storage Store DONEP 10 MG TABLET at room temperature, ideally between 15°C to 30°C (59°F to 86°F). Keep it out of reach of children.

Frequently asked questions

Donepezil 10 mg Tablet, marketed as DONEP 10 MG TABLET by ALKEM LABORATORIES LTD., contains the active ingredient donepezil. It is primarily used to manage symptoms associated with Alzheimer's disease. Below are frequently asked questions about this medication.

What is DONEP 10 MG TABLET used for?

DONEP 10 MG TABLET is used to treat dementia (memory loss and mental changes) associated with mild, moderate, and severe Alzheimer's disease. It helps improve symptoms such as memory, awareness, thinking, communication, and the ability to perform daily activities. It's important to note that this medication does not cure Alzheimer's disease but can help manage its symptoms and may delay the progression of the condition.

How does DONEP 10 MG TABLET work?

DONEP 10 MG TABLET works by increasing the levels of a natural chemical in the brain called acetylcholine. People with Alzheimer's disease typically have lower amounts of acetylcholine, which is crucial for carrying messages between brain cells and plays a key role in memory and thinking. Donepezil, the active ingredient, is an acetylcholinesterase inhibitor, meaning it prevents the enzyme acetylcholinesterase from breaking down acetylcholine. This allows more acetylcholine to accumulate in the brain, enhancing nerve signal transmission and improving cognitive and behavioral symptoms.

What is the usual dosage of DONEP 10 MG TABLET?

The usual starting dose of donepezil is 5 mg, taken once a day in the evening just before bedtime. Your doctor may increase the dose to 10 mg once daily after 4 to 6 weeks, depending on how you respond to the medication and your tolerability. For moderate to severe Alzheimer's disease, doses may sometimes be increased further, up to 23 mg per day, but only after being on a 10 mg daily dose for at least three months and under strict medical supervision. Always follow your doctor's instructions regarding dosage.

What are the possible side effects of DONEP 10 MG TABLET?

Common side effects include nausea, vomiting, diarrhea, loss of appetite, muscle cramps, fatigue, headache, dizziness, trouble sleeping, and abnormal dreams.

Serious side effects that require immediate medical attention include fainting, slow heartbeat, chest pain, new or worsening breathing problems, severe stomach pain or heartburn, black or tarry stools, bloody vomit, seizures, difficulty urinating, or symptoms of a severe allergic reaction such as skin rash, itching, hives, or swelling of the face, lips, tongue, or throat.

Is DONEP 10 MG TABLET safe during pregnancy or breastfeeding?

Pregnancy: Donepezil is classified as a pregnancy category C drug. There is insufficient data on the use of donepezil in pregnant women and its potential to cause developmental abnormalities. It should only be used during pregnancy if the potential benefits justify the potential risks to the fetus. Animal studies have shown some adverse effects on the fetus.

Breastfeeding: It is not known if donepezil or its metabolites pass into breast milk. Therefore, its use during breastfeeding is generally not recommended, or caution is advised, as the effects on a nursing infant are unknown.

Can DONEP 10 MG TABLET be taken with other medications?

DONEP 10 MG TABLET can interact with various other medications. It's important to inform your doctor about all prescription, over-the-counter, and herbal medications you are taking.

Medications to generally avoid include certain antifungal medications (e.g., itraconazole, fluconazole), dextromethorphan/quinidine, dronedarone, pimozide, and thioridazine.

Caution is advised when taken with:

  • Anticholinergic medications: These can reduce the effectiveness of donepezil.
  • Beta-blockers: Concomitant use can increase the risk of a slow heartbeat and low blood pressure.
  • Certain heart rhythm medications: Donepezil can affect heart rhythm, and combining it with other medications that also affect the QT interval may increase risks.
  • Nonsteroidal anti-inflammatory drugs (NSAIDs): These can increase the risk of stomach bleeding and ulcers when taken with donepezil.
  • Muscle relaxants: Donepezil may exaggerate the effects of certain muscle relaxants used during surgery.
  • CYP2D6 and CYP3A4 inhibitors or inducers: These can affect the levels of donepezil in your body, potentially requiring dose adjustments.

What precautions should be taken while taking DONEP 10 MG TABLET?

Before taking DONEP 10 MG TABLET, tell your doctor if you have a history of heart problems (such as slow heartbeats or irregular heart rhythm), lung problems (like asthma or COPD), seizures, stomach ulcers or bleeding, liver or kidney disease, or difficulty urinating (e.g., due to an enlarged prostate).

This medicine may cause dizziness, drowsiness, blurred vision, or muscle cramps, particularly at the start of treatment or when the dose is increased. Exercise caution when driving or operating machinery until you understand how the medication affects you. Alcohol consumption is generally not recommended as it may lessen the effectiveness of donepezil and increase the risk of side effects. Do not stop taking the medication without consulting your doctor, as abrupt discontinuation may worsen symptoms.

What happens if a dose is missed?

If you miss a dose of DONEP 10 MG TABLET, take it as soon as you remember. However, if it is almost time for your next scheduled dose, skip the missed dose and continue with your regular dosing schedule. Do not take a double dose to make up for a missed one. If you miss your doses for seven days or longer, contact your doctor before restarting the medication.

How should DONEP 10 MG TABLET be stored?

DONEP 10 MG TABLET should be stored in a closed container at room temperature, away from excessive heat, moisture, and direct light. Keep the tablets in their original blister pack until it is time to take them to ensure their stability. It is crucial to keep this and all medicines out of the reach of children.
