Description
VONOMAC 10MG Tablet
VONOMAC 10MG Tablet, manufactured by Macleods Pharmaceuticals (MACLEODS-MACPHAR DIV.), is a medication primarily used to treat conditions associated with excessive stomach acid.
Composition
VONOMAC 10MG Tablet contains Vonoprazan 10 mg as its active ingredient. Vonoprazan is a potassium-competitive acid blocker (P-CAB) that works by inhibiting the H+/K+-ATPase enzyme in the stomach's parietal cells, thereby reducing the production of gastric acid. This mechanism allows for faster and more consistent control of gastric pH compared to traditional proton pump inhibitors (PPIs).
Uses
VONOMAC 10MG Tablet is prescribed for a range of acid-related conditions:
- Stomach and Duodenal Ulcers: Treatment and prevention of sores in the lining of the stomach or intestine.
- Gastroesophageal Reflux Disease (GERD) / Reflux Esophagitis: Healing damage to the esophagus caused by excess stomach acid and alleviating associated heartburn and acid reflux.
- Helicobacter pylori (H. pylori) Infection: Used in combination with antibiotics (such as amoxicillin and clarithromycin) to eradicate H. pylori bacteria, which can lead to stomach ulcers.
- Prevention of Ulcer Recurrence: Helps prevent the recurrence of gastric or duodenal ulcers, especially during the use of low-dose aspirin or NSAIDs.
Dosage and Administration
VONOMAC 10MG Tablet can be taken with or without food, as advised by your doctor. It should be swallowed whole with plenty of water.
- General Adult Dosage: The usual starting dose for adults can be 20 mg once daily.
- Reflux Esophagitis (Maintenance Therapy): Typically 10 mg once daily. If efficacy is inadequate, the dosage may be increased up to 20 mg once daily.
- Prevention of Ulcer Recurrence (during low-dose aspirin/NSAID administration): 10 mg once daily.
- H. pylori Eradication (Adjunct): 20 mg twice daily for seven days, as part of a three-drug regimen including amoxicillin and clarithromycin.
Side Effects
Common side effects of VONOMAC 10MG Tablet may include:
- Diarrhea
- Stomach pain
- Nausea
- Indigestion
- Bloating
- High blood pressure
- Urinary tract infection
Serious side effects are rare but may include:
- Kidney problems
- C. difficile-associated diarrhea
- Bone fractures
- Severe skin reactions
- Low vitamin B12 levels
- Low magnesium levels
- Fundic gland polyps (stomach growths)
Consult your healthcare provider if these symptoms persist or worsen.
Precautions
- Allergies: Do not take if allergic to Vonoprazan.
- Liver/Kidney Problems: Use with caution in patients with kidney disease or severe liver problems.
- Pregnancy and Breastfeeding: Limited safety information is available. Consult your doctor if pregnant or breastfeeding.
- Driving: May cause dizziness; avoid driving or operating machinery until you know how the medication affects you.
- Alcohol: Caution is advised when consuming alcohol.
- Drug Interactions: Inform your doctor about all medicines, supplements, or herbals you are taking to avoid potential interactions. VONOMAC may reduce the effectiveness of certain HIV/AIDS medications (e.g., Rilpivirine) and medicines that require an acidic stomach environment to work (e.g., Iron salts, Ketoconazole/Itraconazole).
- Long-term Use: Long-term treatment (more than one year) may require monitoring for deficiencies and may necessitate supplements.
- Other Medical Conditions: Inform your doctor if you have conditions like stomach cancer, inflammation in the kidneys, bacterial diarrhea, bone issues, allergic reactions, or vitamin/mineral deficiencies.
Storage
Store VONOMAC 10MG Tablet at room temperature in a cool and dry place, below 30°C. Keep it away from direct sunlight, moisture, and out of the sight and reach of children and pets.
Frequently asked questions
VONOMAC 10MG TAB, which contains the active ingredient Vonoprazan, is a medication primarily used to reduce stomach acid. It belongs to a class of drugs called potassium-competitive acid blockers (PCABs).
Here are some frequently asked questions about VONOMAC 10MG TAB:
What is VONOMAC 10MG TAB used for?
VONOMAC 10MG TAB is used to treat conditions caused by excessive stomach acid. This includes healing and maintaining the healing of erosive esophagitis (acid-related damage to the esophagus) and relieving associated heartburn. It is also prescribed for heartburn associated with non-erosive gastroesophageal reflux disease (GERD). Additionally, it is used in combination with antibiotics to treat Helicobacter pylori (H. pylori) infection, which can cause stomach ulcers, and to prevent the recurrence of gastric or duodenal ulcers, including those associated with NSAID use.
How does VONOMAC 10MG TAB work?
VONOMAC 10MG TAB works by inhibiting the H+, K+-ATPase enzyme system, often called the "proton pump," located in the parietal cells of the stomach lining. This enzyme is responsible for the final step of acid production. Unlike traditional proton pump inhibitors (PPIs), Vonoprazan binds reversibly and competitively with potassium ions at the pump, suppressing gastric acid secretion. This mechanism allows for rapid and sustained acid suppression without needing acid activation.
What is the usual dosage of VONOMAC 10MG TAB?
The dosage depends on the condition being treated:
- For healing erosive esophagitis and associated heartburn: Typically 20 mg once daily for 8 weeks.
- For maintaining healed erosive esophagitis: Usually 10 mg once daily for up to 6 months.
- For heartburn relief in non-erosive GERD: Generally 10 mg once daily for 4 weeks.
- For H. pylori infection (in combination with antibiotics): 20 mg twice daily, often with amoxicillin and clarithromycin, for 14 days. VONOMAC can be taken with or without food. Your doctor may adjust the dose if you have kidney or liver issues.
What are the possible side effects of VONOMAC 10MG TAB?
Common side effects may include headache, diarrhea, nausea, abdominal pain, indigestion, stomach bloating, and constipation. More serious, though rare, side effects can include severe allergic reactions (e.g., rash, itching, swelling), kidney problems (acute tubulointerstitial nephritis), low magnesium levels (symptoms like muscle cramps or irregular heartbeat), low vitamin B12 levels, and Clostridioides difficile infection (watery stools, stomach pain, fever). Long-term use has also been associated with an increased risk of bone fractures and stomach growths (fundic gland polyps).
Is VONOMAC 10MG TAB safe during pregnancy or breastfeeding?
Pregnancy: There are no sufficient studies on Vonoprazan in pregnant women. Animal studies have shown embryo-fetal toxicity at high doses, and therefore, VONOMAC is generally not recommended during pregnancy unless clearly necessary and the potential benefits outweigh the risks. If used as part of an H. pylori eradication therapy with clarithromycin, it is contraindicated in pregnancy due to risks associated with clarithromycin. Breastfeeding: It is not known if Vonoprazan passes into human milk. However, animal studies suggest excretion into milk and potential liver injury in nursing offspring. The manufacturer advises against breastfeeding during treatment and for 2 days after the last dose due to these potential risks.
Can VONOMAC 10MG TAB be taken with other medications?
Vonoprazan can interact with other medications. It should not be taken with drugs containing rilpivirine (used for HIV-1), as Vonomac can significantly reduce its effectiveness. It may also increase the blood levels of certain drugs like digoxin and can decrease the effectiveness of medications that require stomach acid for absorption, such as nelfinavir, itraconazole, some tyrosine kinase inhibitors (e.g., gefitinib, nilotinib, dasatinib), and iron salts. Vonoprazan can also inhibit the metabolism of some drugs metabolized by cytochrome P450 enzymes. However, it does not appear to have clinically meaningful interactions with low-dose aspirin or certain NSAIDs. Always inform your doctor about all prescription, over-the-counter medications, supplements, and herbal products you are taking.
What precautions should be taken while using VONOMAC 10MG TAB?
Before taking VONOMAC, inform your doctor if you have kidney or liver disease, osteopenia (bone thinning) or osteoporosis, or low levels of calcium, magnesium, or potassium in your blood. Long-term use can lead to conditions like vitamin B12 deficiency and low magnesium levels, requiring periodic monitoring. There's also a risk of stomach polyps with prolonged use. If you experience symptoms like acute kidney inflammation (acute tubulointerstitial nephritis), inform your doctor immediately. Avoid lying down immediately after taking the medication to help prevent acid reflux.
What should I do if I miss a dose of VONOMAC 10MG TAB?
If you miss a dose while taking it once daily (e.g., for erosive esophagitis or non-erosive GERD), take it as soon as you remember, provided it's within 12 hours of your usual time. If more than 12 hours have passed, skip the missed dose and take your next dose at the regularly scheduled time. Do not take a double dose. If you miss a dose while taking it twice daily (e.g., for H. pylori treatment), take it as soon as you remember, provided it's within 4 hours of your usual time. If more than 4 hours have passed, skip the missed dose and take your next dose at the regular time. Do not double doses.
How should VONOMAC 10MG TAB be stored?
VONOMAC 10MG TAB should be stored in a closed container at room temperature, away from excessive heat, moisture, and direct light. It should not be stored above 30°C. Always keep all medicines out of the reach of children.
